摘要:肉桂酸及其酯是一类重要的有机化合物,可作为香精香料、药物中间体、发光材料等,且其本身具有一定的药理活性而备受关注。 本实验以苯甲醛和乙酸酐为原料,K2CO3/γ-Al2O3为催化剂,通过Perkin法合成肉桂酸。考察了不同的催化剂、反应的物料比、温度、回流时间、催化剂的含量、以及相转移催化剂的种类和含量对反应产率的影响,确定出最佳的反应条件为:n(苯甲醛):n (乙酸酐): n (K2CO3) = 1:2:0.5,反应回流温度为150 ℃,回流时间为2 h。测定了产品的熔点,折光率,并对其进行了IR和1H NMR表征。 采用大孔磺酸型树脂D001做催化剂,以肉桂酸和甲醇为反应原料合成肉桂酸甲酯,对酯化反应进行了工艺条件的优化。考察了反应的摩尔比、催化剂用量、反应的回流时间对产率的影响,此外对催化剂的重复利用能力还进行了考察,确定出反应的最佳条件为:n(肉桂酸):n(甲醇)=1:25,催化剂用量25%,反应回流时间为2h。在此较优的条件之下合成肉桂酸乙酯、肉桂酸异丙酯、肉桂酸正丁酯、肉桂酸异戊酯。对合成的产品进行了熔点和折光率测定,并对其进行了IR和1H NMR的表征。 关键词 :苯甲醛,乙酸酐,肉桂酸,肉桂酸酯,K2CO3/γ-Al2O3 ,大孔磺酸树脂D001
Abstract:Cinnamic acid and its esters are an important class of organic compounds. They can be used as flavors and fragrances,pharmaceutical intermediates,and so on.certain pharmacological activity of concern. And they were subject to for concern due to some pharmacological activity of themself. This experiment with benzaldehyde and acetic anhydride as raw materials,K2CO3/ gamma-irradiation Al2O3 as catalyst, through the Perkin synthesis method of cinnamic acid. the effects on the reaction yields such as the type and content of catalysts,the reactants ratios,temperature,reflux time,as well as the type and content of the phase transfer catalyst were investigated . The optimum reaction conditions were: n (benzaldehyde): n (acetic anhydride): n (K2CO3) = 1:2:0.5, reaction temperature 150 ℃, reflux time is 2 h.The melting point and the refractive index were determined,and the products were characterized by 1H NMR and IR. Methyl cinnamate was synthesized by cinnamic acid and methanol,and the sulfonated macroporous resin D001 as catalyst. The conditions on the esterification reaction were optimized. Factors affecting the yield such as the reaction molar ratio,catalyst amount , reflux time were investigated .In addition,the reuse capacity of the catalyst was investigated . To determine the best condition of reaction for: n (cinnamic acid) : n (methanol) = pitted, catalyst 25%, and the reaction backflow time is 2h . Under the better conditions,ethyl cinnamate,isopropyl cinnamate,n-butyl cinnamate,isoamyl cinnamate ester were synthesized. The products were determined by melting point and refractive index,and they were characterized by 1H NMR and IR.
