摘要:以人工胃液或人工肠液模拟体内消化道环境处理不同浓度梯度的低聚糖(低聚木糖、低聚异麦芽糖),再用处理后的不同浓度梯度的低聚糖做为碳源添加到不含碳源的嗜酸乳杆菌基础培养基(MRS培养基)和长双歧杆菌培养基(BBL培养基)中,增殖培养嗜酸乳杆菌和长双歧杆菌。以未经人工胃液及人工肠液处理的低聚糖与葡萄糖作为碳源的培养基作为对照,量化比较体内模拟环境下低聚糖对益生菌的增殖效果。结果显示低聚木糖和低聚异麦芽糖代替葡萄糖为碳源,在体内模拟体外的环境下对嗜酸乳杆菌相较于葡萄糖为碳源具有显著的增殖效果,且低聚木糖增效效果更优。低聚木糖和低聚异麦芽糖代替葡萄糖为碳源在体外模拟体内环境下均对长双歧杆菌有显著的增殖作用。另外我们发现,在体外模拟体内试验与体外试验对比中,低聚糖和葡萄糖两种糖作为碳源情况下,低聚糖浓度越高,其对益生菌的增殖效果越好。且在低聚糖浓度达到100%时,低聚木糖体外模拟体内条件下对嗜酸乳杆菌的增殖效果是体外实验增殖效果的2.25倍。在低聚木糖和低聚异麦芽糖浓度为100%时,两者在体外模拟体内条件下对长双歧杆菌的增殖效果均约为体外试验的两倍。 关键词:低聚木糖 低聚异麦芽糖 嗜酸乳杆菌 长双歧杆菌 益生菌 益生元
Abstract:Dealing with the oligosaccharides of different concentrations of artificial gastric juice orartificial intestinal fluid simulation of in vivo gastrointestinal environment. Proliferationculture of Lactobacillus acidophilus and Bifidobacterium longum by using different concentrations of oligosaccharides and then treated as the carbon sourceadded to the carbon source-free Lactobacillus acidophilus basal medium(MRSmedium) and Bifidobacterium-longum medium (BBL medium).Without the artificial gastric juice and artificial intestinal fluid deal with theoligosaccharides with glucose as the carbon source as ontrols,comparing simulation environment in vivo proliferative effect ofoligosaccharides on probiotic. The results display XOS and Isomaltooligosaccharide of maltose instead of glucose as carbon source in the body to simulate the in vitro environment has a significant multiplier effect of Lactobacillus acidophilus compared with glucose as carbon source, and XOS synergistic effectbetter. XOS and Isomaltooligosaccharide instead of glucose as the carbon source in vitro simulated body environment has a significant role in the proliferation of Bifidobacterium longum. In addition, we found that in vitro simulation of in vivo tests and in vitro test contrast, oligosaccharides and glucose, two sugars as a carbon source case, the oligosaccharide concentration the higher the better probiotics proliferation. And oligosaccharide concentration reached 100%, the effect on the proliferation of Lactobacillus acidophilus in the XOS in vitro under simulated in vivo conditions is 2.25 times the effect of the proliferation of in vitro experiments. In the XOS Isomaltooligosaccharide concentration of 100%, both in the body under the conditions of the in vitro proliferation of Bifidobacterium longum was about twice that of the in vitro tests.
