摘要: 目的:目前己二烯雌酚的市场消耗量逐渐上升,其残留物危害生命健康,研制迅速、简便、廉价的对其检测的方法已迫在眉睫。本实验课题是为研制具有我国自主知识产权的己二烯雌酚残留免疫检测试剂盒奠定基础,主要对己二烯雌酚多克隆抗体抗制备及elisa检测方法等进行了研究。方法:多克隆抗体制备与elisa检测,用人工合成的免疫原de-bsa免疫家兔,采集免疫动物血清。对包被液、封闭液等因素进行优化,应用间接elisa法检测抗血清。其效价为1:6400。结果:通过免疫兔子获得了己二烯雌酚的多克隆抗体,并证明elisa检测的迅速、简便、廉价性。 结论: 抗体表现出对己二烯雌酚较高的特异性和灵敏性。 关键词:己二烯雌酚; 多克隆抗体; ELISA
Abstract: Objective: The current dienestrol market consumption rises gradually, the residues of endangering life and health, develop quickly, simple, cheap for its detection method has been imminent. The experiment is developed with independent intellectual property rights of China dienestrol residual immune detection kit to lay the foundation, mainly on dienestrol polyclonal antibody preparation and determination method of ELISA was studied. Methods: the preparation of polyclonal antibody and ELISA detection, using synthetic immunogenic de-bsa immunization of rabbits, collecting immune animal serum. The coating liquid, closed fluid factors optimization, application of indirect ELISA assay antiserum. The titer of 1:6400. Results: through immune rabbits received dienestrol polyclonal antibody, and prove that ELISA detection of rapid, simple, cheap.
