摘要:本课题主要研究紫甘蓝色素的提取与纯化工艺,以新鲜的紫甘蓝叶为原材料对紫甘蓝色素的提取与纯化条件进行优化,紫甘蓝色素花色苷作为一种天然食用色素,相对于目前食品工业上所用的合成色素,安全、无毒、资源丰富,并且具有一定营养和药理作用,在食品、化妆品、医药品等方面具有巨大的应用潜力。采用常规溶剂提取法提取紫甘蓝中的花色苷,采用大孔树脂吸附法对提取的色素进行纯化,本实验测得紫甘蓝色素花色苷的含量为20.94mg/100g。本文优化了紫甘蓝色素的提取工艺,实验得出提取液的料液比为1﹕50、温度为60 ℃、乙醇浓度为60%、提取时间为60 min、pH为1时,提取效果最好,提取率为40.24%。为了得到含量较高、品质更好的花色苷产品,本文通过大孔吸附树脂的筛选和吸附性能研究,对浸提液进行纯化精制。通过研究4种大孔吸附树脂对紫甘蓝色素吸附率和解吸率,选择NKA树脂作为紫甘蓝色素的纯化树脂。并对吸附条件进行优化,得出在温度30℃时,吸附时间在140-160min之间最适宜,在最佳吸附条件下的吸附率为95.21%;对解吸条件进行优化,得出在75%左右的酒精进行解吸最适宜,解吸的温度50℃最适宜,在最佳解吸收条件下的解吸率为85.47%。 关键词 紫甘蓝;花色苷;提取;纯化;工艺优化
Abstract:This study mainly researched into the technology for separating and purifying pigment from purple cabbage.With fresh purple cabbage leaf as materials,the extraction and purification conditions of purple cabbage pigment were optimized. By conventional solvent the anthocyanin was extracted and by macroporous resin adsorption method the extracting pigment was further purified . In this paper, the process of purple cabbage pigment extraction was optimized. the the best extraction effect was achieved when the ratio of material to liquid was 1:50, and temperature was 60 ℃, ethanol concentration was 60%, extraction time was 60 minand pH was 1.To obtain higher Content and better quality of anthocyanins products we studied the screening and the adsorption performance of macroporous adsorption resin,and finally the extract was purified. After studying the rate of adsorption and desorption of 4 kinds of macroporous adsorption resin, NKA resin was chosen to purify the pigment of purple cabbage. Optimizating the adsorption conditions we concluded that the maximum adsorption amount was abtained at 30 ℃and the most suitable time for the adsorption was 140 - 160min. Optimizating the desorption conditions, we concluded that the most suitable concentration for desorption of alcohol was in about 75%.In this condtion,the adsorption rate is 95.21% and the desorption is 85.47%.
