摘要:本课题通过碱提酸沉法提取黑豆蛋白,采用Alcalase、Neutrase和Flavourzyme蛋白酶水解黑豆蛋白,采用ORAC法测定黑豆蛋白酶解产物及其分离组分的总抗氧化能力,研究不同浓度荧光素钠和AAPH的荧光淬灭曲线,以不同浓度Trolox溶液及谷胱甘肽溶液的荧光淬灭曲线下面的保护面积绘制标准曲线,确定清除氧自由基最佳检测条件,并考察酶标板边际效应的影响以及该方法的重复性、回收率。为黑豆肽的抗氧化活性评价和开发应用提供科学依据。 关键词 黑豆肽;抗氧化能力;ORAC;相关性
Abstract:This issue through alkaline extraction and acid precipitation black bean protein extraction, using Alcalase protease hydrolysis of black beans in Neutrase and Flavourzyme of protein, using the ORAC method for the determination of the total antioxidant capacity of black bean protein hydrolysates and their fractions, the effects of different concentrations of sodium fluorescein and AAPHfluorescence quenching curve, different concentrations of Trolox solution and glutathione solution fluorescence quenching curve and draw the optimum conditions of the standard curve, the marginal effect of the microtiter plates, and the repeatability of the method, the recovery. The the ORAC measurement results with DPPH radical scavenging antioxidant measurement method comparison, analysis of different antioxidant detection method, to provide a scientific basis for the black beans peptide antioxidant activity evaluation and development applications.
