摘要:本文主要采用从土壤中分离到的亚硝化细菌为研究对象,探讨了半连续式、连续式培养对亚硝化细菌的影响,测定了动力学模型相关特征参数。实验结果表明:当采用半连续培养方式,培养40d,每7d更换一次培养基,三角瓶和量筒中亚硝酸盐氮的浓度最高分别达到174.17mg/L和156.33mg/L,采用三角瓶培养的亚硝化细菌脱氮性能优于量筒培养的;当采用连续式培养方式,不断注入新鲜培养基,不断流出原有的培养基,培养56d,亚硝酸盐氮的含量最高可达到202.56mg/L,总体呈现稳定增长的趋势;根据米—门方程和统一动力学方程,测定相关动力学参数,当氨氮含量充足时,菌群数量由开始的迅速增长逐渐趋于平缓,说明此时亚硝化细菌菌群数量的变化与容器的体积有一定的关系;当氨氮含量不足时,菌群数量增长受基质氨氮浓度的影响最大。 关键词:亚硝化细菌;半连续式培养;连续式培养;动力学模型参数
Abstract:This paper mainly uses the nitrite bacteria isolated from soil as the research object, discusses the semi-continuous, continuous culture influence on nitrite bacteria, kinetic model parameters determined. The experimental results show that: when the semi-continuous culture, culture of 40d, each 7d to replace a medium, nitrite nitrogen flask and the cylinder in the highest concentration reached 174.17mg/L and 156.33mg/L, the nitrite bacteria triangular flask culture nitrogen removal performance is better than that of graduate training; when using continuous cultivation, and inject fresh medium, constant flow of medium, the original culture of 56d, the highest content of nitrite nitrogen can reach 202.56mg/L, the overall steady growth trend; according to the Michaelis-Menten equation and unified dynamics equation, determined the kinetic parameters, when the ammonia nitrogen is plentiful, microflora by rapid growth began gradually flatten, description when the number of changes and container nitrosobacteria flora volume has a certain relationship; when the nitrogen content is insufficient, microflora growth affected by ammonia nitrogen concentration maximum matrix.
