摘要:本研究以云芝子实体为原料,将纤维素酶和中性蛋白酶按1:1的比例复合,并作为复合酶来提取云芝子实体,再对云芝提取物进行抗氧化性和抑菌活性研究。研究结果表明,最佳提取工艺为:液料比20:1、复合酶用量2%、温度40℃、酶解时间2h、pH为6.5,在此条件下,云芝提取物的提取率达4.28%。抗氧化实验结果显示:云芝提取物对超氧阴离子自由基具有明显的的清除效果,且清除作用随着提取物浓度的增大而增强;在0.125mg/ml-2.0mg/ml浓度范围内,其清除效果低于VC。抑菌实验结果显示:云芝提取物溶液对大肠杆菌、金黄色葡萄球菌、酵母菌、枯草芽孢杆菌有一定的抑制作用,其最小抑菌浓度(MIC)分别为:0.0125g/ml、0.00625g/ml、0.00625g/ml、0.0125g/ml,而对蜡样芽孢杆菌和黑曲霉无明显抑制作用。 关键词:云芝; 提取物;抗氧化;抑菌性
Abstract:In this paper,polystictus versicolor fruiting bodies was used as raw materials, mixed the cellulase and neutral protease by the ratio of 1:1, and as a composite enzyme to extract polysaccharide from polystictus versicolor , researched its antioxidant and antibacterial activity . The optimal conditions of extract were determined as follows : the liquid to material ratio of 20:1, the amount of enzyme was 2%, temperature of 40℃, reaction time of 2h, pH to 6.5. Under these conditions, the polysaccharides extraction rate reached 4.28%. According to the antioxidant experimental ,we found that polystictus versicolor extract had obvious scavenging effect on superoxide anion radical , but in the concentration range of 0.125mg/ml-2.0mg/ml , its effect was lower than VC . And with increasing of its concentration, the scavenging effect on superoxide anion radical was increased gradually. The antibacterial experimental results showed that: polystictus versicolor extract had antimicrobial effects on Escherichia coli, Staphylococcus aureus, Yeast, Bacillus subtilis, and the minimum inhibitory concentration (MIC) were: 0.0125g/ml, 0.00625g/ml, 0.00625g/ml, 0.0125g/m, but no antimicrobial effects on Bacillus cereus and Aspergillus niger .
