摘要:本实验是以提高组培苗遗传稳定性为目的,进行伴矿景天组培苗茎尖的低温保存初步研究。研究了不同的冷冻保护剂、不同冷冻温度、冷冻保存时间等对伴矿景天组培苗茎尖超低温保存后存活率的影响。结果表明,剪取伴矿景天组培苗的茎尖5-10mm,放入冷冻保护剂(10%DMSO+0.5mol/L葡萄糖或10%甘油+0.5mol/L葡萄糖)处理后,在-10℃或-70℃冷冻保存5d,在37-40℃的水浴中解冻,冻存后的组培苗幼芽用不含琼脂的液体培养基反复冲洗,后转入再生培养基(MS+1.5mg/LNAA+0.1mg/L6-BA+5umol/LCd(NO3)2),暗培养4d,转入光照培养20d,最高存活率分别达到90%或30%。 关键词:组培苗; 冷冻保护剂; 温度; 存活率
Abstract: This paper studied the survival rate of stem tip of tissue culture seeding of Sedum plumbizincicola on different cryoprotectants, temperatures and freezing times. The purpose was to increase the genetic stability of tissue culture seedling. The results showed that clipped 5-10mm from the stem tip of tissue culture seeding of the Sedum plumbizincicola, put in cryoprotectants(10%DMSO+0.5mol/LGlucose,10%DMSO+0.5mol/LSorbitol), cryopreserved for 5 days on -10℃ or -70℃,and thawed in 37℃ to 40 ℃ water bath . The bud of tissue culture seedling washed repeatedly by liquid medium not containing agar. After transfered to the regeneration medium(MS+1.5mg/LNAA+0.1mg/L6-BA+5umol/LCd(NO3)2), dark treatment for 10 days and transfered to light training for 20 days, the highest survival rate could be achieved 90% or 30%.
